Biological Background: PDE3A Function and Localization
- PDE3A (cGMP-inhibited 3',5'-cyclic phosphodiesterase A, CGI-PDE A) is a dual-substrate phosphodiesterase that hydrolyzes both cAMP and cGMP, key second messengers in cellular signaling. Related references: PMID:11420239
- Beyond its catalytic activity, PDE3A participates in an E2/17beta-estradiol-induced pro-apoptotic pathway by stabilizing the PDE3A/SLFN12 complex, leading to SLFN12 dephosphorylation and activation of its rRNA ribonuclease activity.
- The protein is localized to membranes and the cytosol, consistent with its transmembrane domains and roles in compartmentalized cyclic nucleotide signaling.
- PDE3A is a phosphoprotein subject to post-translational modifications, including phosphorylation and ubiquitination, which may regulate its activity and stability.
- It binds metal ions such as magnesium and manganese as cofactors for its phosphodiesterase activity.
- The gene encoding PDE3A is conserved across species, with the mouse protein sharing high homology with human PDE3A (GeneID: 5139).
Experimental Guidance and Technical Tips
- For Western blot, a band around 125 kDa is expected in mouse tissue lysates. Validate the antibody with appropriate positive and negative controls.
- In immunoprecipitation (IP), pre-clearing the lysate and using appropriate negative controls can help reduce non-specific binding.
- For ELISA, titration of both the capture/detection antibody and antigen is recommended to optimize signal-to-noise ratio.
- As a polyclonal antibody, slight batch-to-batch variation is normal; validation in your specific sample system is advised.
