Biological Background: PDCD5 Function and Localization
- PDCD5 (Programmed cell death protein 5), also known as TFAR19 (TF-1 cell apoptosis-related protein 19), is a 14 kDa protein encoded by the PDCD5 gene in humans.
- In response to DNA damage, PDCD5 associates with TP53/p53 to prevent MDM2-mediated degradation of TP53, thereby promoting TP53-mediated signaling and apoptosis. Related references: PMID:19616514, PMID:22914926, PMID:28051100
- PDCD5 also protects KAT5/TIP60 from degradation, further enhancing TP53/p53-mediated signaling. Related references: PMID:19308289
- PDCD5 localizes to the nucleus, cytoplasm, and cytosol, allowing it to interact with cytoplasmic and nuclear partners.
- It is widely expressed across tissues, with highest levels in heart, testis, kidney, pituitary gland, adrenal gland, and placenta.
- Post-translational modifications include acetylation and phosphorylation, which may regulate its function and stability.
- Alternative splicing generates multiple PDCD5 isoforms, potentially modulating its activity.
- As its name implies, PDCD5 is involved in programmed cell death (apoptosis), particularly in the context of DNA damage and TP53 activation.
Experimental Guidance and Technical Tips
- For western blotting (WB), note the predicted molecular weight of ~14 kDa and optimize gel percentage and transfer conditions accordingly.
- For immunofluorescence/ICC, consider appropriate permeabilization and fixation methods to detect PDCD5 in its nuclear and cytoplasmic locations.
- As a polyclonal antibody, validate lot-to-lot consistency and performance in your specific sample system.
- The immunogen spans amino acids 1-125 of human PDCD5; cross-reactivity with mouse and rat is indicated, but verify in your experimental model.
- For ELISA, optimize coating and detection conditions using the recombinant protein immunogen as a positive control if available.
