- PDCD4, also known as H731 or neoplastic transformation inhibitor protein, is a tumor suppressor and apoptosis regulator encoded by the gene PDCD4.
- It inhibits translation initiation and cap-dependent translation by hindering the interaction between EIF4A1 and EIF4G and suppressing EIF4A helicase activity.
- PDCD4 modulates JNK kinase activation and down-regulates MAP4K1, affecting MAPK85 and JUN-dependent transcription, which is important in inhibiting cell invasion.
- Subcellular localization includes both the nucleus and cytoplasm.
- It is up-regulated in proliferative cells and highly expressed in epithelial cells of the mammary gland, with reduced expression in lung cancer and colon carcinoma, supporting its role as a tumor suppressor.
- Post-translational modifications include phosphorylation and acetylation; it is a phosphoprotein and ubiquitin-conjugated protein with repetitive sequences.

Experimental Guidance and Technical Tips
- For Western blot, consider using a reduced sample preparation and validate the band at approximately 52 kDa in lysates from human, mouse, or rat tissues or cell lines.

- For IF/ICC, fix and permeabilize cells optimally; note that PDCD4 localizes to both nucleus and cytoplasm, so expect signal in both compartments.
- Include a positive control such as a cell line with known PDCD4 expression (e.g., proliferative or epithelial cells) and a negative control (e.g., PDCD4 knockdown) to verify antibody performance.
