Biological Background: PD-L1/CD274 Function and Localization
- PD-L1/CD274, also known as B7-H1, is a member of the B7 family of immune regulatory ligands, characterized by immunoglobulin-like domains. It exists as multiple isoforms due to alternative splicing.
- PD-L1 serves as a ligand for the inhibitory receptor PDCD1/PD-1 on T-cells, playing a critical role in negatively regulating T-cell activation and effector functions, which is essential for induction and maintenance of immune tolerance to self. Related references: PMID:11015443, PMID:28813410, PMID:28813417
- Through an unidentified activating receptor, PD-L1 may costimulate T-cell subsets that predominantly produce interleukin-10 (IL10). Related references: PMID:10581077
- Under hypoxic conditions, PD-L1 can translocate to the nucleus and act as a transcription coactivator by interacting with phosphorylated STAT3, promoting GSDMC transcription and leading to pyroptosis. Related references: PMID:32929201
- The PD-1/PD-L1 axis is often exploited by tumors to inhibit cytotoxic T lymphocyte (CTL) responses, facilitating immune escape. Blockade of this interaction can restore anti-tumor immunity, making it a key target for cancer immunotherapy. Related references: PMID:28813410, PMID:28813417
- PD-L1 is predominantly localized to the cell membrane, but can also be found in early endosomes, recycling endosomes, and is secreted in soluble form. Additionally, it can translocate to the nucleus under specific conditions.
- PD-L1 is widely expressed, with high levels in heart, skeletal muscle, placenta, and lung. It is also expressed on activated T-cells, B-cells, dendritic cells, keratinocytes, monocytes, and various peripheral blood cells.
Experimental Guidance and Technical Tips
- This antibody was generated against recombinant protein corresponding to amino acids 1-290 of human PD-L1 (Q9NZQ7), covering the extracellular domain.
- For Western blot (WB), a band around 33 kDa is expected; consider using validated positive controls such as PD-L1-expressing cell lines (e.g., human tumor lines) or tissues like placenta or lung.
- For ELISA applications, optimize coating and detection conditions using the recombinant immunogen as a standard.
- Due to the soluble form and potential nuclear localization, fractionation studies may require careful lysis buffer selection.
