Biological Background: PCSK9 Function and Localization

• Proprotein convertase subtilisin/kexin type 9 (PCSK9), also known as NARC-1 or PC9, is a secreted serine protease that belongs to the proprotein convertase family. It is synthesized as a zymogen and undergoes autocatalytic cleavage in the endoplasmic reticulum.
• PCSK9 is a crucial regulator of plasma cholesterol homeostasis by binding to low-density lipoprotein (LDL) receptor family members (LDLR, VLDLR, LRP1, LRP8) and promoting their intracellular degradation, thereby reducing LDL cholesterol uptake.
• It enhances hepatic LDLR degradation via a clathrin-dependent pathway, likely by preventing recycling of LDLR from endosomes to the cell surface or directing it to lysosomes, and can also induce LDLR ubiquitination.
• Beyond LDLR, PCSK9 inhibits intracellular degradation of apolipoprotein B (APOB) through an LDLR-independent autophagosome/lysosome pathway and participates in the disposal of non-acetylated BACE1 intermediates in the early secretory pathway.

• The immunogen covers amino acids 35–694 of mouse PCSK9, spanning the catalytic domain and C-terminal region; the antibody may recognize both the full-length pro-form (~74 kDa) and the mature cleaved form. Consider validating detected bands with appropriate controls.
• For Western blotting, use a reducing SDS-PAGE and anticipate a major band near 74 kDa in mouse liver or hepatocyte lysates; the antibody cross-reacts with human and rat, but confirm reactivity in your target species.
• In IF/ICC experiments, PCSK9 displays a punctate cytoplasmic and perinuclear distribution consistent with ER, Golgi, and endosomal compartments. Permeabilization is required; optimize fixation and antibody dilution for your cell type.
