Biological Background: PCSK9 Function and Localization

• PCSK9 (Proprotein convertase subtilisin/kexin type 9) is a crucial regulator of plasma cholesterol homeostasis by binding to LDL receptor family members (LDLR, VLDLR, LRP1, LRP8) and promoting their degradation in intracellular acidic compartments. Related references: PMID:18039658

• For Western blot, note that PCSK9 can be detected as a full-length ~74 kDa protein and a ~20 kDa fragment, likely corresponding to the cleaved prodomain. Additional bands may appear depending on sample preparation and endogenous cleavage. Confirm band patterns using appropriate controls.

• For immunofluorescence and immunoprecipitation, select an antibody dilution compatible with your protocol and cell type; include positive and negative controls to assess specificity.
