Biological Background: PCSK9 Function and Localization

  • PCSK9 is a proprotein convertase subtilisin/kexin type 9 (also known as NARC-1, PC9) that plays a central role in cholesterol homeostasis by binding to LDL receptor family members and promoting their degradation (PubMed:18039658). Related references: PMID:18039658
  • It enhances hepatic LDLR degradation via a clathrin-mediated pathway, likely by preventing recycling from endosomes or directing to lysosomes, and can induce LDLR ubiquitination (PubMed:17461796, PubMed:18197702, PubMed:18799458). Related references: PMID:17461796 PMID:18197702 PMID:18799458
  • PCSK9 also inhibits APOB degradation via the autophagosome/lysosome pathway independently of LDLR, and is involved in disposal of BACE1 intermediates in the secretory pathway (PubMed:18660751). Related references: PMID:18660751
  • It regulates neuronal apoptosis by modulating LRP8/APOER2 levels and associated anti-apoptotic signaling pathways.
  • Subcellularly, PCSK9 localizes to the cytoplasm, secreted, endosome, lysosome, cell surface, endoplasmic reticulum, and Golgi apparatus.
  • Tissue expression includes neuro-epithelioma, colon carcinoma, hepatic and pancreatic cell lines, and Schwann cells.
  • Key features include autocatalytic cleavage, alternative splicing, extensive glycosylation, phosphorylation, and sulfation; mutations are linked to familial hypercholesterolemia and hypocholesterolemia.

Experimental Guidance and Technical Tips

  • This antibody is generated using a recombinant protein immunogen spanning amino acids 31-692 of human PCSK9, covering the catalytic and C-terminal domains, which may contribute to pan-PCSK9 recognition.
  • For Western blotting, PCSK9 is detected as a ~74 kDa proprotein and a ~20 kDa cleaved product; consider optimizing sample preparation to visualize both forms.
  • The antibody is validated for IHC-P, IF/ICC, and ELISA; validate in the relevant sample system (e.g., liver tissue for cholesterol metabolism studies) with appropriate controls.
  • Cross-reactivity with mouse PCSK9 is reported; verify sequence homology for other species if needed.
