Biological Background: PCMT1 Function and Localization
- PCMT1 (Protein-L-isoaspartate(D-aspartate) O-methyltransferase), also known as PIMT, is encoded by the PCMT1 gene (UniProt P22061) and functions as a repair methyltransferase.
- It initiates the repair of damaged proteins by catalyzing methyl esterification of abnormal L-isoaspartyl and D-aspartyl residues that accumulate spontaneously in aging peptides and proteins. Related references: PMID:3167043, PMID:6469980
- Known substrates include EIF4EBP2, microtubule-associated protein 2, calreticulin, clathrin light chains, ubiquitin C-terminal hydrolase isozyme L1, phosphatidylethanolamine-binding protein 1, stathmin, and synucleins (by similarity).
- Localizes predominantly to the cytosol, consistent with its role in repairing intracellular proteins.
- Belongs to the methyltransferase superfamily, utilizing S-adenosyl-L-methionine as a methyl donor; the enzyme is also subject to acetylation and undergoes alternative splicing.
- The calculated molecular weight of the unmodified protein is approximately 24.6 kDa; the antibody detects an endogenous band around 25 kDa.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 62–286 of human PCMT1, a conserved region; the antibody is expected to recognize full‐length PCMT1 as well as isoforms sharing this epitope.
- For western blot (WB), consider using a standard reducing SDS-PAGE protocol and validate detection in your target species; reported reactivity includes human, mouse, and rat.
- In immunofluorescence/immunocytochemistry (IF/ICC), optimize fixation and permeabilization conditions as signal may be influenced by cellular context; include appropriate controls.
- This polyclonal antibody is suitable for immunoprecipitation (IP) and ELISA; however, individual assay conditions (e.g., antibody dilution, incubation time) should be empirically determined.
