Biological Background: PCIF1 Function and Localization
- PCIF1 (Phosphorylated CTD-interacting factor 1), also known as CAPAM (Cap-specific adenosine methyltransferase), hCAPAM, hPCIF1, C20orf67, or PPP1R121, is an mRNA (2'-O-methyladenosine-N6-)-methyltransferase.
- It catalyzes the formation of N6,2'-O-dimethyladenosine cap (m6A(m)) on the second transcribed nucleotide of capped mRNAs (PubMed:30467178, PubMed:30487554, PubMed:31279658).
Related references: PMID:30467178, PMID:30487554, PMID:31279658 - PCIF1 is recruited to the RNA polymerase II (RNAPII) early elongation complex through interaction with POLR2A, enabling co-transcriptional m6A(m) modification.
- Subcellularly, PCIF1 localizes to the nucleus, and it is ubiquitously expressed across tissues as indicated by proteomics data.
- It belongs to the methyltransferase family and is annotated as a reference proteome component and phosphoprotein (UniProt Keywords).
Experimental Guidance and Technical Tips
- The antibody was raised against a recombinant protein spanning the N-terminal region (amino acids 1-200) of human PCIF1. This region may contain functional domains relevant to its methyltransferase activity.
- For Western blot (WB), a band around 81 kDa is predicted; however, actual migration may vary. Validate specificity by using appropriate positive and negative controls (e.g., PCIF1-overexpressing or knockdown cell lysates).
- The antibody is reported to cross-react with mouse and rat PCIF1, making it suitable for studies in these model organisms. Confirm cross-reactivity in your specific samples.
- For immunohistochemistry (IHC-P), optimize antigen retrieval and antibody dilution. Consider including a blocking peptide or negative control tissue to assess specificity.
- For ELISA, the recombinant immunogen can be employed as a positive control to establish assay conditions.
