Biological Background: PCBD1 Function and Localization
- PCBD1 (Pterin-4-alpha-carbinolamine dehydratase), also known as DCoH, PCD, PHS, or PCBD, is a bifunctional protein with enzymatic and transcriptional coactivator activities.
- It plays a critical role in tetrahydrobiopterin (BH4) biosynthesis by preventing the formation of 7-pterins and accelerating the formation of quinonoid-BH2 (By similarity).
- As a dimerization cofactor of HNF1A (DCoH), PCBD1 regulates the dimerization of homeodomain protein HNF1A and enhances its transcriptional activity (By similarity).
- PCBD1 also acts as a coactivator for HNF1B-dependent transcription (PubMed:24204001). Related references: PMID:24204001
- The protein localizes to both the cytoplasm and nucleus, consistent with its dual roles in metabolism and transcription regulation.
