Biological Background: PCBD1 Function and Localization

  • PCBD1 (Pterin-4-alpha-carbinolamine dehydratase), also known as DCoH, PCD, PHS, or PCBD, is a bifunctional protein with enzymatic and transcriptional coactivator activities.
  • It plays a critical role in tetrahydrobiopterin (BH4) biosynthesis by preventing the formation of 7-pterins and accelerating the formation of quinonoid-BH2 (By similarity).
  • As a dimerization cofactor of HNF1A (DCoH), PCBD1 regulates the dimerization of homeodomain protein HNF1A and enhances its transcriptional activity (By similarity).
  • PCBD1 also acts as a coactivator for HNF1B-dependent transcription (PubMed:24204001). Related references: PMID:24204001
  • The protein localizes to both the cytoplasm and nucleus, consistent with its dual roles in metabolism and transcription regulation.
  • Key post-translational modification includes acetylation; it is classified as a lyase and a transcriptional activator.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide derived from the N-terminal region (aa 1-104) of human PCBD1. This region is highly conserved, suggesting potential reactivity with human, mouse, and rat PCBD1.
  • For Western blotting (WB), the expected band size is ~12 kDa. Depending on the sample, both cytoplasmic and nuclear fractions may be analyzed, and loading controls should be appropriate to each fraction.
  • In immunohistochemistry (IHC-P), antigen retrieval methods and dilution should be optimized for the specific tissue type; both nuclear and cytoplasmic staining may be observed.
  • For ELISA, the antibody is expected to capture or detect both recombinant and endogenous PCBD1; users should validate sensitivity and specificity in their own assay systems.
  • Due to the presence of acetylation modifications, the antibody's binding may be affected by the modification status of the target; consider this when interpreting results.
  • Cross-reactivity with mouse and rat is supported by sequence homology; however, users should verify specificity in their experimental models.
