Biological Background: PAX5 Function and Localization
- PAX5 (Paired box protein Pax-5), also known as B-cell-specific transcription factor (BSAP), is a transcription factor critical for B-lymphocyte lineage commitment. It functions as both a repressor of B-lineage inappropriate genes and an activator of B-lineage-specific genes, thereby directing lymphoid progenitors to the B-cell fate. Related references: PMID:10811620, PMID:27181361
- PAX5 regulates downstream processes essential for B-cell development, including cell adhesion and migration, induction of V(H)-to-D(H)J(H) recombination, facilitation of pre-B-cell receptor signaling, and promotion of maturation to the mature B-cell stage. Related references: PMID:32612238
- Within pro-B cells, PAX5 represses the cohesin-release factor WAPL, leading to global changes in chromosomal architecture that facilitate the generation of a diverse antibody repertoire. Related references: PMID:32612238
- PAX5 plays a role in Epstein-Barr virus (EBV) infection by maintaining EBV genome copy number through promotion of EBNA1/oriP-dependent binding and transcription, and by inhibiting lytic EBV reactivation via modulation of BZLF1 activity. Related references: PMID:31941781, PMID:23678172
- Subcellularly, PAX5 localizes predominantly in the nucleus.
- Additional keywords associated with PAX5 highlight its involvement in developmental processes, differentiation, neurogenesis, and spermatogenesis; its DNA-binding paired box domain; and its classification as a proto-oncogene due to its role in chromosomal rearrangements linked to disease.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 150-300 of human PAX5, a region that may include part of the paired domain. Polyclonal antibodies raised against this segment are likely to recognize multiple epitopes, providing robust detection in applications such as Western blotting and immunoprecipitation.
- For Western blotting, expect a primary band near 42kDa in human B-cell lysates. However, validation in the specific experimental system is recommended, as splice variants or post-translational modifications could alter the apparent molecular weight.
- This antibody has been validated for use in WB, IP, and ELISA. When performing immunoprecipitation, consider using a cross-linking strategy to immobilize the antibody and minimize heavy/light chain interference in subsequent detection.
- Cross-reactivity is reported for human only. If detection of PAX5 from other species is required, validate the antibody in the relevant sample system with appropriate positive and negative controls.
