Biological Background: POZ-, AT hook-, and zinc finger-containing protein 1 (PATZ1) Function and Localization
- PATZ1 is a transcriptional regulator featuring a BTB/POZ domain, AT hook, and multiple C2H2-type zinc fingers. It participates in embryogenesis, senescence, T-cell development, and neurogenesis. Mechanistically, it interacts with TP53/p53 to control genes involved in proliferation and DNA-damage response by inhibiting p53's DNA binding and transcriptional activity. - During microbial infection, PATZ1 plays a positive role in viral cDNA synthesis.
- Subcellular localization is primarily nuclear.
- It is ubiquitously expressed across tissues.
- Post-translational modifications include phosphorylation and ubiquitination (isopeptide bond formation), and the protein undergoes alternative splicing. Chromosomal rearrangements involving PATZ1 are associated with proto-oncogenic activity.
- The molecular weight of PATZ1 is approximately 74 kDa.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 250–350 of human PATZ1. Given high sequence conservation, the antibody is expected to cross-react with mouse and rat PATZ1 (as indicated by cross-reactivity data).
- For Western blotting, consider optimizing antibody dilution and incubation conditions for your specific sample type. A band near 74 kDa is expected.
- The antibody is validated for ELISA; perform titration experiments to determine the optimal working concentration for your assay.
