Biological Background: PASK Function and Localization

  • PASK is a PAS domain-containing serine/threonine-protein kinase encoded by the gene PASK (KIAA0135) in humans. It is also known as STK37 or PASKIN.
  • It functions as a sensor of environmental conditions such as oxygen, glucose, and nutrition, playing a role in energy homeostasis and protein translation regulation.
  • PASK phosphorylates key substrates including EEF1A1 (translation elongation factor), GYS1 (glycogen synthase), PDX1 (pancreatic and duodenal homeobox 1), and RPS6 (ribosomal protein S6), thereby modulating glycogen synthesis, insulin production, and translation efficiency.
  • The kinase is localized in both the cytoplasm and nucleus, consistent with its regulatory roles.
  • PASK is ubiquitously expressed, with relatively higher levels in brain, prostate, and testis; at the protein level, it is present in germ cells of testis and the midpiece of sperm tails.

  • This polyclonal antibody is suitable for Western blot (WB), immunohistochemistry (paraffin) (IHC-P), immunofluorescence/immunocytochemistry (IF/ICC), and ELISA. Optimize dilutions and conditions based on your experimental system.
  • For WB, a band at approximately 143 kDa is expected. Consider using a broad-range marker and appropriate gel percentage to resolve this high molecular weight protein.
  • Cross-reactivity has been validated with human, mouse, and rat samples. For other species, check sequence homology of the immunogen region.
