Biological Background: PARP2 Function and Localization
-
Poly-ADP-ribosyltransferase that mediates poly-ADP-ribosylation of proteins and plays a key role in DNA repair. Related references: PMID:10364231 PMID:25043379 PMID:27471034
-
Mediates glutamate, aspartate or serine ADP-ribosylation of proteins; serine ADP-ribosylation constitutes the primary form in response to DNA damage. Related references: PMID:25043379 PMID:30104678 PMID:30321391 PMID:32939087
-
Initiates repair of double-strand DNA breaks by recognizing DNA breaks, recruiting HPF1, and licensing serine ADP-ribosylation of histones to promote chromatin decompaction and repair factor recruitment. Related references: PMID:10364231 PMID:32939087 PMID:34108479
-
HPF1 interaction confers serine specificity and restricts polymerase activity to limit poly-ADP-ribose chain length. Related references: PMID:28190768 PMID:32028527 PMID:34108479 PMID:34732825 PMID:34795260
-
Specifically mediates formation of branched poly-ADP-ribosylation, which is recognized by factors such as APLF. Related references: PMID:30104678
-
Also able to ADP-ribosylate DNA, preferentially acting on 5'-terminal phosphates at strand break termini in nicked duplex. Related references: PMID:27471034 PMID:29361132
-
Localizes to the nucleus and chromosomes.
-
Widely expressed, mainly in actively dividing tissues; highest levels in brain, heart, pancreas, skeletal muscle and testis. Related references: PMID:10364231
Experimental Guidance and Technical Tips
- The immunogen peptide (aa 484-583) corresponds to a region in the C-terminal half of PARP2; the antibody may recognize epitopes within this fragment. Consider validating specificity with full-length recombinant protein or knockout cell lines.
- For western blot, a band of approximately 66 kDa is expected in human cell lysates. Use appropriate controls to confirm detection of endogenous PARP2.
- In ELISA applications, this antibody can be used for direct detection; however, optimization of coating antigen and antibody concentration is recommended for your specific assay format.
- Cross-reactivity with other PARP family members (e.g., PARP1) should be evaluated, especially in applications requiring high specificity.
