Biological Background: PARL Function and Localization

  • PARL (Presenilin-associated rhomboid-like protein, mitochondrial) is a mitochondrial intramembrane cleaving protease, also known as PSARL, RHBDS1, or PRO2207.
  • It is required for apoptosis control during postnatal growth, essential for proteolytic processing of anti-apoptotic OPA1, which prevents mitochondrial cytochrome c release in response to intrinsic apoptotic signals (by similarity).
  • Mediates maturation of PINK1 into its 52kDa form after cleavage by MPP (PubMed:22354088). Related references: PMID:22354088
  • Promotes cleavage of PGAM5 in damaged mitochondria in response to mitochondrial membrane potential loss (PubMed:22915595). Also mediates differential cleavage of PINK1 and PGAM5 depending on mitochondrial health status (PubMed:22915595). Related references: PMID:22915595
