Biological Background: PARG Function and Localization
- Encodes poly(ADP-ribose) glycohydrolase, which hydrolyzes the ribose-ribose bonds in poly(ADP-ribose) polymers. Related references: PMID:15450800, PMID:21892188, PMID:23102699
- Acts primarily as an exo-glycohydrolase and can also function as an endo-glycohydrolase at low efficiency, releasing poly(ADP-ribose) fragments and ADP-ribose monomers. Related references: PMID:23102699, PMID:23481255, PMID:37268618
- Unable to cleave the ester bond between terminal ADP-ribose and ADP-ribosylated residues, leaving mono-ADP-ribosylated proteins. Related references: PMID:21892188, PMID:23474714, PMID:33186521
- Rapidly degrades poly(ADP-ribose) synthesized after DNA damage, preventing detrimental accumulation during prolonged replicative stress. Related references: PMID:23102699, PMID:34019811, PMID:24906880
- Required for retinoid acid-dependent gene transactivation by removing poly(ADP-ribose) from histone demethylase KDM4D, facilitating chromatin derepression. Related references: PMID:23102699
- Participates in nuclear ATP generation together with PARP1, NMNAT1, and NUDT5, supporting energy-consuming chromatin remodeling. Related references: PMID:27257257
- Localizes to the nucleus, cytoplasm, and mitochondrion matrix.
- Ubiquitously expressed across tissues.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 777-976 of human PARG, a C-terminal region that includes part of the catalytic domain; consider this when designing epitope-specific assays.
- For Western blot, a band at approximately 111 kDa is expected; verify using appropriate positive controls such as HeLa or Jurkat cell lysates.
- In ELISA applications, use recombinant human PARG protein as a coating antigen and optimize antibody dilution to achieve optimal signal-to-noise ratio.
