Biological Background: PAR4 Function and Localization

• Also known as Prostate apoptosis response 4 protein (Par-4), PAR4 is a pro-apoptotic regulator encoded by the PAWR gene in humans.
• PAR4 selectively induces apoptosis in cancer cells while sparing normal cells, sensitizing them to various apoptotic stimuli and promoting tumor regression in animal models.
• It triggers apoptosis via activation of the Fas death receptor pathway and concurrent inhibition of NF-kappa-B transcriptional activity.
• PAR4 interacts with WT1 to inhibit transcriptional activation and enhance repression, leading to down-regulation of the anti-apoptotic protein BCL2.
• It may also function directly as a transcriptional repressor and is implicated in regulating BACE1-mediated cleavage of the amyloid precursor protein (APP).
