Biological Background: PAR2 Function and Localization
- PAR2, also known as F2RL1, GPR11, Coagulation factor II receptor-like 1, and Thrombin receptor-like 1, is a G protein-coupled receptor encoded by the F2RL1 gene.
- Functions as a receptor for trypsin and trypsin-like enzymes, activating downstream signaling pathways such as PLC, MAPK, I-kappaB kinase/NF-kappaB, and Rho. Related references: PMID:28445455
- Plays a key role in modulating inflammatory responses and innate/adaptive immunity, acting as a sensor for proteolytic enzymes during infection; can signal synergistically with TLR4 and modulate TLR3 signaling.
- Contributes to endothelial barrier integrity: mediating barrier protective signaling via coagulation factor Xa, and regulating barrier integrity during neutrophil extravasation via PRTN3 cleavage. Related references: PMID:22409427 PMID:23202369
- Proposed to have a bronchoprotective role in airway epithelium, but can also compromise the epithelial barrier by interrupting E-cadherin adhesion. Related references: PMID:10086357
- Associates with G proteins alpha subunits including GNAQ, GNA11, GNA12, GNA13, and can additionally signal through G(i) subunit alpha. Related references: PMID:21627585
- Subcellular localization is primarily at the cell membrane.
- Tissue expression: widely expressed, with highest levels in pancreas, liver, kidney, small intestine, and colon; not detected in brain or skeletal muscle. Related references: PMID:7556175 PMID:8615752
Experimental Guidance and Technical Tips
- The immunogen spans the N-terminal region (aa 15–94) of human PAR2; the antibody may recognize epitopes within this domain. Consider validating cross-reactivity in your species of interest.
- For WB, a band around 44 kDa is expected. Take into account potential glycosylation that may shift the observed molecular weight.
- In IF/ICC, verify staining specificity using appropriate positive and negative control cell lines or knockdown models.
- For ELISA, the antibody can be tested as a capture or detection reagent in your assay system; optimal pairings should be determined experimentally.
