Biological Background: PAR2 Function and Localization
- Proteinase-activated receptor 2 (PAR2), also known as coagulation factor II receptor-like 1 (F2RL1), G protein-coupled receptor 11 (GPR11), and thrombin receptor-like 1, is a member of the G protein-coupled receptor superfamily.
- Acts as a receptor for trypsin and trypsin-like enzymes, coupling to G proteins to activate phospholipase C, intracellular calcium, MAPK, and NF-kappaB pathways.

- Involved in cellular migration, cytoskeletal rearrangement, and chemotaxis through beta-arrestin-promoted scaffolds, promoting cofilin dephosphorylation and actin filament severing.
- Widely expressed with high levels in pancreas, liver, kidney, small intestine, and colon; expressed in endothelial cells.

Experimental Guidance and Technical Tips
- The immunogen covers amino acids 298–397 in the intracellular C‑terminal tail of human PAR2, making the antibody suitable for detecting total PAR2 in denaturing (WB) and immunohistochemical (IHC-P) applications.
- For WB, run appropriate molecular weight markers and expect a band near 44 kDa; consider using a blocking peptide or knockout/knockdown lysate as a negative control.
- In IHC-P, optimize antigen retrieval for your samples and validate staining patterns in tissues with known PAR2 expression (e.g., pancreas, colon).
- For ELISA, pairing with a detection antibody that recognizes a non-overlapping epitope may improve signal specificity.
