Biological Background: PAPSS1 Function and Localization
- PAPSS1 is a bifunctional enzyme that mediates two sequential steps in the sulfate activation pathway: first, ATP sulfurylase activity generates adenosine 5′-phosphosulfate (APS) from ATP and sulfate; second, APS kinase activity phosphorylates APS to produce 3′-phosphoadenylylsulfate (PAPS), the universal sulfate donor.

- PAPSS1 is required for the biosynthesis of sulfated L-selectin ligands in endothelial cells, which are critical for lymphocyte homing. Related references: PMID:9576487
- The protein is widely expressed, with highest levels in testis, pancreas, kidney, thymus, prostate, ovary, small intestine, colon, leukocytes, liver, and high endothelial venules.

- For Western blot, expect a band around 71 kDa; consider using reducing conditions and validate with appropriate positive controls (e.g., HeLa or HEK293 cell lysates).
- In ELISA, the antibody may be suitable for both direct and indirect detection; titration and antigen coating optimization are recommended.
- Because PAPSS1 is ubiquitously expressed, include a panel of tissue/cell extracts to assess specificity.
