Biological Background: Terminal nucleotidyltransferase 4A (PAPD7/TENT4A) Function and Localization
- Encodes a terminal nucleotidyltransferase, also known as TENT4A, PAPD7, POLS, TRF4-1, and LAK-1.
- Catalyzes the transfer of ATP and GTP onto RNA 3' poly(A) tails, creating heterogeneous poly(A) tails that stabilize mRNAs by protecting them from active deadenylation. Related references: PMID:23376078, PMID:30026317
- Functions as the catalytic subunit of a TRAMP-like complex, adding short poly(A) tails to nuclear RNAs to target them for exosome-mediated degradation.
- Lacks terminal uridylyltransferase activity and does not participate in replication-dependent histone mRNA degradation.
- Localizes to both the cytoplasm and the nucleus (nucleoplasm).
- Contains ATP- and metal-binding sites; subject to alternative splicing.

- The immunogen corresponds to amino acids 303–542 of human PAPD7; homology across species supports the reported cross-reactivity with human, mouse, and rat.
- For Western blot, a band around 85 kDa is expected; consider using lysates from cell lines or tissues expressing PAPD7 as positive controls.
- For IHC-P, perform antigen retrieval and validate staining specificity in appropriate formalin-fixed, paraffin-embedded tissues.
