Biological Background: Pannexin-1 Function and Localization
- Pannexin-1 (PANX1) is an integral membrane protein and a member of the pannexin family, forming hexameric channels at the plasma membrane and endoplasmic reticulum.
- Functions as an anion-selective channel with relatively low conductance, playing key roles in blood pressure regulation, apoptotic cell clearance, and oogenesis. Related references: PMID:15304325 PMID:16908669 PMID:20829356
- Upon phosphorylation or C-terminal cleavage, mediates ATP and UTP release, acting as a large-pore channel that facilitates communication and phagocyte attraction during apoptosis. Related references: PMID:32238926
- May function as a Ca(2+)-leak channel in the endoplasmic reticulum membrane, contributing to ER calcium homeostasis. Related references: PMID:16908669
- Widely expressed with highest levels in oocytes and brain, and low levels in sperm cells. Related references: PMID:30918116
- Subject to alternative splicing and post-translational modifications including phosphorylation, S-nitrosylation, and N-glycosylation.
- Disease-associated aliases (OOMD7, OZEMA7) link PANX1 to oculodentodigital dysplasia and related disorders.
- Contains four transmembrane helices with disulfide bonds stabilizing structure; predicted molecular weight ~48 kDa.
Experimental Guidance and Technical Tips
- The immunogen used is a synthetic peptide corresponding to amino acids 50-150 of human Pannexin-1 (NP_056183.2). This region may include an extracellular or intracellular loop; consider optimization of antibody incubation conditions for native vs. denatured protein detection.
- For Western blot, consider a starting dilution of 1:500-1:2000, and validate with appropriate positive and negative controls in your sample system.
- For ELISA, this antibody can be used for direct antigen detection or as part of a sandwich pair; we recommend cross-testing with a matched detection antibody for optimal performance.
- Note that Pannexin-1 is expressed in many tissues and may undergo cleavage-induced activation in apoptotic samples; take precautions to minimize post-lysis modifications if studying channel function.
