Biological Background: Pantothenate kinase 2, mitochondrial Function and Localization
- Pantothenate kinase 2, mitochondrial (PANK2) catalyzes the first and rate-limiting step in coenzyme A (CoA) biosynthesis: the ATP-dependent phosphorylation of pantothenate to 4'-phosphopantothenate. Related references: PMID:15659606, PMID:16272150, PMID:17242360
- Alternative splicing and initiation produce both a mitochondrial isoform (containing a transit peptide) and a cytoplasmic isoform; the mitochondrial form additionally localizes to the nucleus.
- PANK2 is required for the angiogenic activity of human umbilical vein endothelial cells (HUVEC). Related references: PMID:30221726
- The protein is ubiquitously expressed with highest levels in testis and brain, consistent with its role in CoA-dependent metabolic and neurological processes.
- Mutations in PANK2 are linked to pantothenate kinase-associated neurodegeneration (PKAN), a subtype of neurodegeneration with brain iron accumulation (NBIA).
- Classified as a kinase, transferase, and phosphoprotein, PANK2 is an ATP-binding enzyme central to cellular metabolism.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a central region (aa 300-400) of human PANK2; verify target detection in the relevant species (human, mouse) and sample type.
- For Western blot, the predicted molecular weight is 63 kDa; consider using reducing conditions and include appropriate positive controls (e.g., HeLa, HEK293, or mouse testis lysates).
- Cross-reactivity with mouse is reported; validate specificity in mouse models if applicable.
- For ELISA, use recombinant protein or peptide to confirm antibody binding and optimize coating conditions.
