Biological Background: pan-PKC Function and Localization
- pan-PKC antibody recognizes multiple protein kinase C (PKC) isoforms: PRKCA, PRKCB, PRKCD, PRKCE, PRKCH, and PRKCQ, which belong to the AGC serine/threonine kinase family.
- PKC alpha (PRKCA, SWISS: P17252) is a calcium-activated, phospholipid- and diacylglycerol (DAG)-dependent kinase that regulates cell proliferation, apoptosis, differentiation, migration, adhesion, and tumorigenesis.
- PRKCA phosphorylates key substrates such as RAF1 (activating MAPK/ERK signaling), BCL2 (anti-apoptotic), and TNNT2/cardiac troponin T (reducing myocardial contractility).
- In chemokine-induced T cell migration, PRKCA phosphorylates DOCK8, leading to its dissociation from LRCH1 and activation of CDC42 (PubMed:28028151). Related references: PMID:28028151
- PRKCA is found in the cytoplasm, cell membrane, mitochondrial membrane, and nucleus, with dynamic subcellular localization depending on activation status.
- This kinase is implicated in angiogenesis, platelet aggregation, and cardiac hypertrophy, and is often overexpressed in cancers such as gliomas and breast carcinomas.

Experimental Guidance and Technical Tips
- The immunogen sequence (aa 572–672 of human PRKCA) lies in a conserved C-terminal region, which may confer cross-reactivity among PKC isoforms; validate detection of each target isoform in your sample system.
- For Western blot, expect bands in the range of 59–83 kDa (depending on the isoform); include appropriate positive and negative controls to confirm specificity.
- In IF/ICC, permeabilization can affect PKC staining patterns; consider using 0.1% Triton X-100 or digitonin, and note that subcellular distribution (cytoplasm, membrane, nucleus) may vary with cell type and stimulation.
- For ELISA, titrate the antibody to determine the optimal working concentration for your assay conditions.
