Biological Background: PALD1 Function and Localization
- Paladin (PALD1, also known as PALD and KIAA1274) is a human protein with a predicted molecular weight of approximately 97 kDa.
- Subcellular localization studies indicate that paladin is predominantly found in the cytoplasm and cytosol.
- At the tissue level, paladin is expressed in endothelial cells and mural cells of certain larger vessels, and possibly in brain microglia. Protein expression has been confirmed in peripheral blood mononuclear cells.
- UniProt keywords annotate paladin as a lipoprotein with myristate and phosphoprotein modifications, suggesting that myristoylation and phosphorylation may regulate its function.
- Although the precise biological role of paladin is not yet fully defined, it is included in the human reference proteome and has been identified in proteomics screens.
Experimental Guidance and Technical Tips
- This polyclonal antibody was raised against an N-terminal recombinant fragment (aa 1–300) and is expected to recognize full-length paladin.
- For Western blotting, a specific band of approximately 97 kDa may be detected; consider including positive and negative controls and validating in human, mouse, or rat cell lysates.
- In immunofluorescence/ICC, optimize fixation and permeabilization conditions to preserve cytoplasmic localization.
- The antibody is also suitable for ELISA applications; titration experiments are recommended to determine optimal working concentrations.
- Always validate the antibody in your specific experimental system before drawing conclusions.
