Biological Background: PAK6 Function and Localization

• PAK6 (also known as PAK-5 or p21-activated kinase 6) is a serine/threonine-protein kinase that plays a role in the regulation of gene transcription. It inhibits androgen receptor (AR)- and estrogen receptor (ESR1)-mediated transcription by phosphorylating the DNA-binding domain of AR.
• PAK6 may also regulate the cytoskeleton by interacting with IQGAP1, and it can protect cells from apoptosis through phosphorylation of BAD.
• The kinase activity of PAK6 is induced by various effectors, including AR and MAP2K6/MAPKK6.
• PAK6 is an ATP-binding kinase member of the p21-activated kinase (PAK) family, and it undergoes alternative splicing, producing multiple isoforms.
• Subcellular localization is both cytoplasmic and nuclear.
• PAK6 is selectively expressed in brain and testis, with lower levels detected in other tissues such as prostate and breast.
