PAK2 (also known as PAK65, PAKgamma, p58, PAK-2, S6/H4 kinase) is a serine/threonine kinase that acts as a downstream effector of the small GTPases CDC42 and RAC1. - For Western blotting, detect full-length PAK2 at ~58 kDa; under apoptotic conditions, a 34 kDa caspase-cleaved fragment may appear. Include appropriate positive controls.
- In immunofluorescence/ICC, anticipate cytoplasmic and nuclear staining with possible perinuclear enrichment. Optimize fixation and permeabilization for each cell type.
- For ELISA, titrate against the immunogen peptide (aa 1–100) to confirm specificity; cross-reactivity to mouse and rat suggests broad mammalian utility.
- When using mouse or rat samples, validate reactivity with species-specific positive controls, as cross-reactivity is reported but not quantified.
