Biological Background: PAK2 Function and Localization

  • PAK2 (also known as PAK65, PAKgamma, p58, PAK-2, S6/H4 kinase) is a serine/threonine kinase that acts as a downstream effector of the small GTPases CDC42 and RAC1.   • Full-length PAK2 promotes cell survival and growth, and phosphorylates JUN to mediate EGF-induced proliferation. It also phosphorylates MAPK4 and MAPK6, activating MAPKAPK5 to regulate F-actin polymerization and cell migration. Related references: PMID:7744004, PMID:21177766, PMID:21317288

  • PAK2 phosphorylates numerous substrates including histone H4 to facilitate nucleosome assembly of H3.3-H4, BAD, ribosomal protein S6, and caspase-7 (CASP7), inhibiting CASP7 activity. Related references: PMID:21724829, PMID:21555521, PMID:27889207

  • In immunofluorescence/ICC, anticipate cytoplasmic and nuclear staining with possible perinuclear enrichment. Optimize fixation and permeabilization for each cell type.
  • For ELISA, titrate against the immunogen peptide (aa 1–100) to confirm specificity; cross-reactivity to mouse and rat suggests broad mammalian utility.
  • When using mouse or rat samples, validate reactivity with species-specific positive controls, as cross-reactivity is reported but not quantified.
