Biological Background: PAI-1/Serpin E1 Function and Localization

  • PAI-1, also known as Serpin E1, is encoded by the SERPINE1 gene and belongs to the serine protease inhibitor (serpin) superfamily.
  • It is a primary inhibitor of tissue-type plasminogen activator (PLAT) and urokinase-type plasminogen activator (PLAU), playing a key role in down-regulating fibrinolysis and controlling blood clot degradation. Related references: PMID:17912461, PMID:8481516, PMID:9207454
  • By inhibiting PLAU, PAI-1 regulates cell adhesion and spreading, and it also acts as a regulator of cell migration independently of its protease inhibitor activity. Related references: PMID:9175705, PMID:15001579, PMID:9168821
  • The protein is required for keratinocyte migration during cutaneous wound repair and is involved in cellular and replicative senescence. Related references: PMID:18386027, PMID:16862142

Experimental Guidance and Technical Tips

  • For western blotting, a band of approximately 45 kDa is expected under reducing conditions. Use endothelial cell lysates or plasma as positive controls.
  • In immunofluorescence and immunocytochemistry, consider permeabilization to detect intracellular pools of this secreted protein. Validate in relevant cell types such as HUVECs.
  • The immunogen corresponds to amino acids 24–199; verify sequence conservation if using samples from species other than human, mouse, or rat.
  • For ELISA, pairing with a suitable capture antibody may enhance sensitivity; titrate the antibody to optimize signal-to-noise ratio.
