Biological Background: PAF1 Function and Localization
- Core component of the PAF1 complex (PAF1C), which regulates transcription elongation by RNA polymerase II, histone modifications, and mRNA 3' end formation.
- PAF1C associates with RNA polymerase II CTD in both non-phosphorylated and phosphorylated states, facilitating coupling between transcription and co-transcriptional processes.
- Promotes histone H2B K120 monoubiquitination (H2BK120ub1) via recruitment of the RNF20/40 ubiquitin ligase complex, and stimulates H3K4me3 methylation; these modifications are critical for active transcription.
- Required for transcription of Hox and Wnt target genes, and plays a role in hematopoiesis and leukemogenesis through interactions with KMT2A/MLL1 fusion oncoproteins.
- Localizes to the nucleus, consistent with its transcriptional regulatory functions.
- Exhibits oncogenic activity; implicated in alternative splicing, phosphorylation, and ubiquitin conjugation pathways.
- During influenza A (H3N2) infection, interacts with viral NS1 protein to modulate host gene transcription.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the N-terminal region (aa 1-100) of human PAF1, thus the antibody may detect full-length and certain isoforms. Verify isoform specificity in your experimental system.
- For Western blot, expect a band near 60 kDa under reducing conditions; include positive controls such as HeLa or HEK293 cell lysates.
- IHC-P staining should show nuclear localization; optimize antigen retrieval and antibody dilution using appropriate positive tissue controls.
- In ELISA, the peptide immunogen can serve as a standard. Cross-reactivity with mouse and rat PAF1 allows application across these species; confirm performance in the relevant species’ samples.
- Consider co‑immunoprecipitation or ChIP assays if the antibody functions in native protein complex analysis, but validate those applications independently.
