  • Glycodelin (PAEP), also known as placental protein 14 (PP14), pregnancy-associated endometrial alpha-2 globulin (PAEG), and zona-binding inhibitory factor-1 (ZIF-1), is a secreted glycoprotein. Four glycoforms—Glycodelin-S, -A, -F, and -C—exhibit distinct glycosylation and biological roles: Glycodelin-A has contraceptive and immunosuppressive activities, Glycodelin-C stimulates sperm-zona binding, Glycodelin-F inhibits sperm-zona binding and the acrosome reaction, and Glycodelin-S maintains sperm in an uncapacitated state.   • Glycodelin is the major secretory product of the endometrium during the mid-luteal phase and early pregnancy. It exhibits tissue-specific glycoform distribution: Glycodelin-A in amniotic fluid, endometrium/decidua, and maternal serum; Glycodelin-F in follicular fluid, luteinized granulosa cells, and oviduct; Glycodelin-S in seminal plasma and seminal vesicles; and Glycodelin-C in cumulus cells, where it is taken up and glycan-remodeled from Glycodelin-A and -F.   • For WB, consider using reducing SDS-PAGE and a positive control such as human endometrial tissue lysate or seminal plasma, as Glycodelin is highly expressed in these tissues. Due to glycosylation, the observed band may appear slightly higher than the predicted 21 kDa. Optimize primary antibody dilution (e.g., 1:500-1:2000) based on sample abundance.
  • For ELISA, perform checkerboard titrations to determine the optimal coating concentration and antibody dilution. The antibody may be used for direct or indirect detection formats.
  • Cross-reactivity is reported for human only; it is not expected to detect mouse or rat Glycodelin. Validate species specificity in the relevant assay system if needed.
