Biological Background: PADI4 Function and Localization
- PADI4 (Protein-arginine deiminase type-4) is a calcium-dependent hydrolase that catalyzes the citrullination (deimination) of arginine residues in target proteins, playing a key role in post-translational modification and chromatin regulation.
- It citrullinates histones: H1 at Arg-54 (H1R54ci), H3 at Arg-2/8/17/26 (H3R2ci, H3R8ci, H3R17ci, H3R26ci), and H4 at Arg-3 (H4R3ci), thereby modulating nucleosome dynamics and gene expression.
Related references: PMID:15339660 PMID:15345777 PMID:16567635 - In stem cells, PADI4-mediated citrullination of H1R54ci displaces histone H1 from chromatin, inducing global chromatin decondensation and promoting pluripotency maintenance.
Related references: PMID:15339660 PMID:15345777 PMID:16567635 - During innate immune responses in neutrophils, PADI4 drives profound chromatin decondensation and is essential for the formation of neutrophil extracellular traps (NETs), which trap and eliminate pathogens.
Related references: PMID:18209087 PMID:19153223 PMID:25622091 PMID:28574339 - Citrullination of histone H3 by PADI4 prevents its methylation by the arginine methyltransferases CARM1 and PRMT1, leading to transcriptional repression.
Related references: PMID:15345777 - Beyond histones, PADI4 also citrullinates non-histone substrates such as the acetyltransferase EP300 (p300) at Arg-2142, which enhances its interaction with the coactivator NCOA2.
Related references: PMID:15731352 - PADI4 localizes to the cytoplasm, nucleus, and cytoplasmic granules, consistent with its roles in both cytosolic and nuclear processes.
- Its expression is primarily restricted to eosinophils and neutrophils; it is not detected in peripheral monocytes or lymphocytes.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 290–663, encompassing the C-terminal catalytic domain; the antibody is expected to detect full-length PADI4.
- For Western blotting (WB), the predicted molecular weight is 74 kDa; consider using neutrophil cell lines (e.g., HL-60) or primary granulocytes as positive controls.
- In immunofluorescence (IF/ICC), expect a cytoplasmic, nuclear, and granular staining pattern; permeabilization and fixation protocols may need optimization.
- For ELISA applications, validate the antibody pair with recombinant PADI4 protein or cell lysates to determine optimal coating and detection conditions.
