Biological Background: PABPC4 Function and Localization
- PABPC4 (Polyadenylate-binding protein 4), also known as activated-platelet protein 1 (APP-1) or inducible poly(A)-binding protein (iPABP), belongs to the poly(A)-binding protein family.
- It binds specifically to the poly(A) tail of mRNA, a key step in mRNA stabilization and translation initiation. Related references: PMID:8524242
- PABPC4 interacts with SMIM26 mRNA and modulates its post-transcriptional regulation, demonstrating its role in specific mRNA metabolism. Related references: PMID:37009826
- It may play a broader role in cytoplasmic regulatory processes of mRNA metabolism, potentially binding non-poly(A) RNA sequences in vivo.
- The protein is primarily localized in the cytoplasm.
- PABPC4 is expressed at low levels in resting normal T cells, but its mRNA levels are rapidly up-regulated upon T-cell activation, indicating a role in immune response regulation.
- Post-translational modifications include methylation, phosphorylation, isopeptide bond formation, and ubiquitin-like conjugation (Ubl), which may regulate its activity and stability.
- The protein has a molecular weight of approximately 70.8 kDa (UniProt) / 71 kDa (observed).
Experimental Guidance and Technical Tips
- For Western blot, a band at approximately 71 kDa is expected; validate using appropriate positive controls (e.g., activated T-cell lysates).
- For IHC-P, consider antigen retrieval conditions; cytoplasmic staining is anticipated.
- For IF/ICC, permeabilization may be necessary to access the cytoplasmic PABPC4.
- Due to its polyclonal nature, this antibody can be suitable for ELISA as a detection antibody.
- Cross-reactivity with mouse and rat suggests sequence conservation; validate in the relevant sample system.
