Biological Background: p75NTR/NGFR Function and Localization
- Encodes tumor necrosis factor receptor superfamily member 16 (TNFRSF16), also known as low-affinity nerve growth factor receptor (LNGFR) or p75NTR.
- Serves as a low-affinity receptor for mature neurotrophins (NGF, BDNF, NTF3, NTF4) and forms a high-affinity heterodimeric receptor with SORCS2 for precursor neurotrophins. Related references: PMID:24908487
- Plays critical roles in neuronal fate determination, capable of mediating both cell survival and apoptosis, and inactivates RHOA. Related references: PMID:26646181
- Regulates insulin-dependent glucose uptake by modulating GLUT4 translocation through RAB31 in adipocytes and skeletal muscle (By similarity).
- Required for circadian rhythm maintenance by regulating clock gene expression (BMAL1, PER1, PER2, NR1D1) in the suprachiasmatic nucleus and liver, and influences metabolic gene regulation. Related references: PMID:23785138
- Together with BFAR, negatively regulates NF-kappa-B and JNK-related pro-inflammatory signaling pathways. Related references: PMID:22566094
- Subcellularly localizes to the cell membrane, cytoplasm, perikaryon, growth cones, and dendritic spines, reflecting its diverse signaling functions.
- Post-translational modifications include glycosylation, phosphorylation, and disulfide bond formation; alternative splicing yields isoforms such as soluble p75ICD.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 300–400 within the intracellular domain, suggesting recognition of full-length and possibly truncated p75ICD isoforms; validate specificity using isoform-specific controls.
- For western blot, anticipate a band near 45 kDa; confirm identity with positive controls (e.g., neuronal cell lines) and negative controls (e.g., p75NTR knockout lysates if available).
- In IHC-P, optimal antigen retrieval and detection may require titrating antibody concentration; evaluate using known p75NTR-expressing tissues (e.g., basal forebrain, Schwann cells).
- For ELISA, test binding to recombinant p75NTR extracellular domain or peptide to assess affinity; include cross-reactivity checks with related TNFRSF members.
- Cross-reacts with human, mouse, and rat; verify species-specific reactivity in your experimental system and consider blocking conditions to minimize non-specific binding.
