Biological Background: p75NTR Function and Localization
- Encodes TNFRSF16 (tumor necrosis factor receptor superfamily member 16), also known as p75NTR, a low-affinity neurotrophin receptor that binds NGF, BDNF, NTF3, and NTF4.
- Forms a heterodimeric receptor with SORCS2 that binds precursor forms of NGF, BDNF, and NTF3 with high affinity, while showing much lower affinity for mature NGF and BDNF. Related references: PMID:24908487
- Plays a critical role in differentiation and survival of specific neuronal populations during development, and can mediate both cell survival and cell death of neural cells.
- Localizes to the cell membrane, cytoplasm, perikaryon, growth cones, and dendritic spines, consistent with its functions in neuronal signaling.
- Regulates the inactivation of RHOA, a small GTPase involved in cytoskeletal dynamics. Related references: PMID:26646181
- Necessary for the circadian oscillation of clock genes (BMAL1, PER1, PER2, NR1D1) in the suprachiasmatic nucleus and liver, and of genes involved in glucose and lipid metabolism in the liver. Related references: PMID:23785138
- Together with BFAR, negatively regulates NF-kappa-B and JNK-related signaling pathways, influencing apoptosis and stress responses. Related references: PMID:22566094
- Contains structural features including a signal peptide, transmembrane helix, disulfide bonds, and glycosylation sites, with a calculated molecular weight of approximately 45 kDa.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 301-400 of human p75NTR/NGFR (NP_002498.1). For immunoassays, consider peptide blocking to confirm signal specificity, especially when using WB or ELISA.
- This antibody has been reported to cross-react with human, mouse, and rat. Ensure that the target epitope is conserved in your species of interest by aligning the immunogen sequence.
- Western blot using this polyclonal antibody may detect a band around 45 kDa, corresponding to the full-length glycosylated form; however, alternative splicing or post-translational modifications may yield additional bands. Validate with appropriate controls.
- For ELISA applications, test a range of antibody dilutions to determine optimal sensitivity and signal-to-noise ratio in your sample matrix.
