Biological Background: p75NTR/NGFR Function and Localization
-
Encoded by the NGFR gene, p75NTR/NGFR (CD271/TNFRSF16) is a single-pass type I membrane glycoprotein and member of the tumor necrosis factor receptor superfamily. It is also known as low-affinity nerve growth factor receptor (LNGFR) or Gp80-LNGFR.
-
Acts as a low-affinity receptor for mature neurotrophins NGF, BDNF, NTF3, and NTF4, and forms a high-affinity heterodimeric receptor with SORCS2 for precursor forms of NGF, BDNF, and NTF3. Related references: PMID:24908487
-
Regulates diverse neuronal functions including differentiation, survival, and programmed cell death of specific neuronal populations during development. Signaling can promote either survival or apoptosis depending on cellular context.
-
Plays a role in the inactivation of RHOA, contributing to cytoskeletal dynamics. Related references: PMID:26646181
-
Involved in insulin-responsive GLUT4 translocation to the cell surface in adipocytes and skeletal muscle cells, thereby regulating glucose uptake.
-
Required for circadian oscillation of clock genes (BMAL1, PER1, PER2, NR1D1) in the suprachiasmatic nucleus and liver, and modulates hepatic glucose and lipid metabolism genes. Related references: PMID:23785138
-
Together with BFAR, negatively regulates NF-kappa-B and JNK signaling pathways, influencing inflammatory and stress responses. Related references: PMID:22566094
-
Subcellularly, p75NTR/NGFR localizes to the cell membrane, cytoplasm, perikaryon, growth cones, and dendritic spines. It undergoes N-glycosylation and phosphorylation, and exists in multiple alternatively spliced isoforms.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 300–400 within the intracellular domain; the antibody may perform optimally in applications where the epitope is accessible (e.g., denaturing WB).
- For western blot, consider using positive control lysates from human, mouse, or rat neuronal tissues or cell lines (e.g., SH-SY5Y, PC12) expressing p75NTR/NGFR.
- In IHC-P, antigen retrieval conditions should be optimized; the antibody is expected to detect both full-length and truncated isoforms.
- For ELISA, peptide-based assays or recombinant protein capture may be considered; validate with appropriate standards.
- Cross-reactivity with mouse and rat is reported, but potential species-specific differences in epitope recognition should be verified experimentally.
