Biological Background: p63 Function and Localization
- p63, also known as Tumor protein 63, is a member of the p53 family of transcription factors, with multiple isoforms generated by alternative promoter usage and alternative splicing, including TA and DeltaN isoforms that differ in their N-terminal transactivation domains.
- Functions as a sequence-specific DNA-binding transcriptional activator or repressor, regulating genes involved in epithelial morphogenesis, apoptosis, and development. Isoform 2 activates RIPK4 transcription.
- Plays a critical role in limb formation from the apical ectodermal ridge and is required for proper epithelial stratification and maintenance of epithelial stem cell compartments.
- Involved in Notch signaling by probably inducing JAG1 and JAG2, and may be required in conjunction with TP73/p73 for initiation of TP53-dependent apoptosis in response to genotoxic stress.
- Binds to an upstream regulatory region of the DSG1 gene and promotes its transcription, contributing to epithelial differentiation. Related references: PMID:24594668
- Localizes predominantly to the nucleus, consistent with its role as a transcription factor.
- Widely expressed in tissues including heart, kidney, placenta, prostate, skeletal muscle, testis, and thymus, with tissue-specific isoform expression. DeltaN isoforms are highly expressed in progenitor cell layers of skin, breast, eye, and prostate. Isoform 10 is predominantly found in skin squamous cell carcinomas.
