Biological Background: p63 Function and Localization
- p63 (TP63) is a member of the p53 transcription factor family, with multiple isoforms generated by alternative promoter usage and alternative splicing, including transactivating (TA) and N-terminally truncated (DeltaN) variants.
- Functions as a sequence-specific DNA-binding transcriptional activator or repressor; isoform-specific activity is determined by the presence of transactivation and auto-regulatory domains.
- Plays a critical role in epithelial morphogenesis and stratification; the ratio of DeltaN and TA isoforms regulates maintenance of epithelial stem cell compartments and initiation of stratification from embryonic ectoderm.
- Involved in Notch signaling, likely by inducing JAG1 and JAG2, and participates in p53-dependent apoptosis when cooperating with TP73/p73 in response to genotoxic stress or oncogene activation.
- Known target genes include CDKN1A (p21) and DSG1; it promotes DSG1 transcription by binding to its upstream regulatory region. Related references: PMID:24594668
- Localizes to the nucleus; widely expressed in tissues such as heart, kidney, placenta, prostate, skeletal muscle, testis, and thymus, with high DeltaN-type expression in progenitor cell layers of skin, breast, eye, and prostate.
