Biological Background: p63 (Tumor protein 63) Function and Localization
- p63 (Tumor protein 63) is a member of the p53 family, also known as p40, p51, chronic ulcerative stomatitis protein (CUSP), and keratinocyte transcription factor KET. It produces multiple isoforms through alternative promoter usage and splicing, which contain varying transactivation and inhibiting domains.
- Acts as a sequence-specific DNA-binding transcriptional activator or repressor involved in the regulation of epithelial morphogenesis, stem cell maintenance, and limb formation from the apical ectodermal ridge.
- Required, in conjunction with TP73, for initiation of p53/TP53-dependent apoptosis in response to genotoxic insults and activated oncogenes.
- Involved in Notch signaling by probably inducing JAG1 and JAG2.
- Activates transcription of the p21 promoter and binds to an upstream regulatory region of the DSG1 gene to promote its transcription.

- Predominantly localized to the nucleus.
- Widely expressed, notably in heart, kidney, placenta, prostate, skeletal muscle, testis, and thymus. DeltaN-type isoforms are highly expressed in progenitor cell layers of skin, breast, eye, and prostate. Isoform 10 is predominantly expressed in skin squamous cell carcinomas but not in normal skin tissues.
- Contains zinc-finger DNA-binding domains, is post-translationally modified by isopeptide bond formation, and is implicated in developmental processes, ectodermal dysplasia, and premature ovarian failure.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant protein covering amino acids 1-160 of human p63 (NP_003713.3), a region that may be present in multiple isoforms. Consider confirming isoform specificity using isoform-specific detection methods if needed.
- For Western blot, the predicted molecular weight of p63 isoforms ranges from 45-77 kDa; expect multiple bands due to alternative splicing and promoter usage. Use appropriate positive controls (e.g., cell lines or tissues known to express p63).
- For immunofluorescence/ICC, p63 localizes to the nucleus. Validate nuclear staining by counterstaining with a nuclear marker and compare with appropriate negative controls.
- Cross-reactivity is reported for human, mouse, and rat, but verification in your target species and sample system is recommended.
