Biological Background: CHAF1B Function and Localization
- Encoded by the CHAF1B gene, the protein is also known as Chromatin assembly factor 1 subunit B, CAF-I 60 kDa subunit (CAF-I p60), and M-phase phosphoprotein 7.
- Functions as a core component of the Chromatin Assembly Factor 1 (CAF-1) complex, a histone chaperone that mediates the first step of nucleosome assembly by depositing newly synthesized histones H3 and H4 onto replicating DNA.
- Essential for DNA replication-coupled chromatin assembly and DNA repair, ensuring proper packaging of newly synthesized DNA into chromatin.
- The CAF-1 complex is conserved across eukaryotes and plays a critical role in maintaining epigenetic information during cell division.
- Subcellular localization is primarily nuclear, consistent with its role in DNA replication and repair; however, it can also shuttle to the cytoplasm.
- Contains WD repeats that mediate protein-protein interactions within the CAF-1 complex and with other chromatin factors.
- Post-translational modifications include acetylation and phosphorylation (notably during M phase), which may regulate its activity and interactions.
- Acts as a phosphoprotein, with phosphorylation potentially modulating its function during the cell cycle.
Experimental Guidance and Technical Tips
- The antibody was raised against amino acids 459-559 of human p60 CAF1. Confirm that this epitope is present in your target species and isoform.
- For Western blot, use appropriate loading controls and validate the antibody in your specific sample lysate; the predicted molecular weight is approximately 61 kDa.
- For immunohistochemistry (paraffin-embedded sections), antigen retrieval may be required; optimization of retrieval buffer and conditions is recommended.
- In ELISA, coat the antigen under appropriate conditions and consider using a blocking buffer to reduce nonspecific binding.
- Cross-reactivity has been validated for human; if using in other species, sequence homology should be checked and validation performed.
- Always include positive and negative controls to confirm specificity.
