Biological Background: p53 Function and Localization
- p53 (TP53 gene) is a multifunctional transcription factor and tumor suppressor, inducing cell cycle arrest, DNA repair, or apoptosis in response to cellular stress. Related references: PMID:11025664 PMID:12524540 PMID:12810724
- It negatively regulates cell division by controlling the expression of a set of genes, including an inhibitor of cyclin-dependent kinases. Related references: PMID:11025664 PMID:12524540 PMID:12810724
- Apoptosis induction is mediated through stimulation of BAX and FAS antigen expression, repression of Bcl-2, and interactions with ASPP family proteins (PPP1R13B/ASPP1, TP53BP2/ASPP2, PPP1R13L/iASPP). Related references: PMID:12524540 PMID:17189187
- Multiple isoforms (2, 4, 7) exist with distinct regulatory effects on transactivation and apoptosis, enhancing or suppressing isoform 1 functions.
- Subcellular localization includes nucleus, cytoplasm, mitochondria, endoplasmic reticulum, centrosome, and PML bodies, reflecting its diverse roles.
- Ubiquitous tissue expression with isoform-specific patterns; some isoforms are absent in specific tissues like brain, lung, or prostate.
- Post-translational modifications include phosphorylation, acetylation, methylation, and ubiquitination, regulating its stability and activity.
- p53 regulates the circadian clock by repressing CLOCK-BMAL1-mediated transcriptional activation of PER2. Related references: PMID:24051492
Experimental Guidance and Technical Tips
- The immunogen was a synthetic peptide from amino acids 218-390 of human p53; for Western blotting, consider testing primary antibody dilutions starting at 1:500–1:2000, followed by standard detection methods.
- For immunoprecipitation, validate antibody-antigen binding under non-denaturing conditions with appropriate buffers; capture efficiency may require optimization.
- In ELISA, the antibody can serve as a detection or capture reagent; check compatibility with your plate format and detection system.
- As a polyclonal antibody, lot-to-lot variability is expected—validate each new batch in your relevant sample system.
- The antibody is raised against human p53; cross-reactivity with other species has not been determined; if using non-human samples, verify reactivity experimentally.
