- Tumor protein p53-inducible nuclear protein 1 (TP53INP1), also known as p53DINP1 or SIP, is a stress-induced protein with antiproliferative and proapoptotic functions.
- Acts as a dual regulator of transcription and autophagy; positively regulates autophagy by interacting with autophagosome-associated proteins such as GABARAP, GABARAPL1/L2, and MAP1LC3A/B/C.
- Plays a major role in p53/TP53-driven oxidative stress response, possessing both p53-independent antioxidant activity and p53-dependent transcription regulatory functions.
- Stimulates the apoptotic and cell cycle regulatory capacities of p53 and p73; promotes p53 phosphorylation on Ser-46 after double-strand DNA breaks to induce apoptosis.
- Functions as a tumor suppressor by inducing cell death via autophagy- and caspase-dependent mechanisms, and can reduce cell migration by regulating SPARC expression.
- Localizes to the cytoplasm, cytosol, nucleus (including PML bodies), and autophagosomes, reflecting its dynamic role in stress response and autophagy.
