Biological Background: p53 DINP1 Function and Localization

  • Tumor protein p53-inducible nuclear protein 1 (TP53INP1), also known as p53DINP1 or SIP, is a stress-induced protein with antiproliferative and proapoptotic functions.
  • Acts as a dual regulator of transcription and autophagy; positively regulates autophagy by interacting with autophagosome-associated proteins such as GABARAP, GABARAPL1/L2, and MAP1LC3A/B/C.
  • Plays a major role in p53/TP53-driven oxidative stress response, possessing both p53-independent antioxidant activity and p53-dependent transcription regulatory functions.
  • Stimulates the apoptotic and cell cycle regulatory capacities of p53 and p73; promotes p53 phosphorylation on Ser-46 after double-strand DNA breaks to induce apoptosis.
  • Functions as a tumor suppressor by inducing cell death via autophagy- and caspase-dependent mechanisms, and can reduce cell migration by regulating SPARC expression.
  • Localizes to the cytoplasm, cytosol, nucleus (including PML bodies), and autophagosomes, reflecting its dynamic role in stress response and autophagy.
  • Ubiquitously expressed across tissues and identified as a proteomics target; belongs to a gene family with alternative splicing variants.

Experimental Guidance and Technical Tips

  • The immunogen used is a synthetic peptide from the central region (aa 100-200) of human p53 DINP1; consider sequence alignment with mouse and rat orthologs to confirm epitope conservation across species.
  • For Western blot, a band around 27 kDa is expected; validate in positive control cell lines treated with stress inducers to enhance TP53INP1 expression.
  • In IHC-P and IF/ICC, optimize antigen retrieval and permeabilization conditions, as the protein can localize to both nuclear and cytoplasmic compartments.
  • For ELISA, ensure the coating antigen contains the immunogen sequence to maintain detection sensitivity.
