Biological Background: P4HB (Protein Disulfide-Isomerase) Function and Localization

  • P4HB encodes protein disulfide-isomerase (PDI), also known as PDIA1, ERBA2L, and other aliases. It is a multifunctional enzyme belonging to the protein disulfide isomerase family.
  • PDI catalyzes the formation, breakage, and rearrangement of disulfide bonds. At the cell surface, it functions as a reductase that cleaves disulfide bonds of attached proteins, potentially modifying exofacial proteins. Intracellularly, it ensures correct disulfide bond formation in nascent proteins.
  • At high concentrations and following phosphorylation by FAM20C, PDI acts as a chaperone that inhibits aggregation of misfolded proteins. Conversely, at low concentrations, it facilitates aggregation (anti-chaperone activity). Related references: PMID:32149426

  • For Western blot (WB), expect a band at approximately 57 kDa. Validate the antibody in your specific sample system, as post-translational modifications may cause minor shifts.
  • In immunoprecipitation (IP) and immunofluorescence (IF/ICC), native protein conformation may be recognized; optimize lysis and fixation conditions to preserve epitopes.
