Biological Background: P4HB Function and Localization

  • P4HB encodes Protein disulfide-isomerase (PDI), a multifunctional endoplasmic reticulum (ER) chaperone and oxidoreductase that catalyzes disulfide bond formation, breakage, and rearrangement in nascent proteins.

  • P4HB serves as a structural subunit of several multimeric enzymes, including prolyl 4-hydroxylase (involved in collagen synthesis) and microsomal triacylglycerol transfer protein (MTTP), highlighting its role in diverse biosynthetic pathways.
