Biological Background: P4HB Function and Localization
- P4HB encodes Protein disulfide-isomerase (PDI), a multifunctional endoplasmic reticulum (ER) chaperone and oxidoreductase that catalyzes disulfide bond formation, breakage, and rearrangement in nascent proteins.
- It also functions as a cell-surface reductase, cleaving exofacial protein disulfide bonds and modulating plasma membrane redox state, particularly in Th2 cells via interaction with LGALS9. Related references: PMID:21670307
- P4HB serves as a structural subunit of several multimeric enzymes, including prolyl 4-hydroxylase (involved in collagen synthesis) and microsomal triacylglycerol transfer protein (MTTP), highlighting its role in diverse biosynthetic pathways.
- At high concentrations and upon phosphorylation by FAM20C, P4HB acts as a chaperone to inhibit misfolded protein aggregation, while at low concentrations it facilitates aggregation, demonstrating concentration-dependent anti-chaperone activity. Related references: PMID:32149426

- For Western blotting, a major band at approximately 57 kDa is expected in human, mouse, and rat samples; consider using reduced denaturing conditions.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), validate the antibody with appropriate positive and negative control tissues/cells, as P4HB is abundantly expressed in the ER.
