Biological Background: p47phox/NCF1 Function and Localization

  • p47phox (Neutrophil cytosol factor 1) is encoded by the NCF1 gene (also known as NOXO2, SH3PXD1A) and is a 47 kDa cytosolic protein; alternative splicing has been reported but not fully characterized.
  • As a subunit of the phagocyte NADPH oxidase complex, p47phox enables electron transfer from cytosolic NADPH to O2 to produce superoxide anion (O2(-)). Related references: PMID:2547247, PMID:2550933, PMID:38355798
  • Activation involves phosphorylation-dependent binding of p47phox to the C-terminus of CYBA/p22-phox, initiating assembly of cytosolic subunits with the core membrane complex. Related references: PMID:12732142, PMID:19801500
  • p47phox is primarily located in the cytoplasm and cytosol, and translocates to the membrane upon activation.
  • Detected at the protein level in peripheral blood monocytes and neutrophils.
  • Features include SH3 domains, a PX domain for lipid binding, and phosphorylation sites, consistent with its regulatory role.
  • Defects in NCF1 are associated with autosomal recessive chronic granulomatous disease (CGD), characterized by impaired phagocytic killing.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to residues 291–390 of human p47phox; if cross-reactivity to other species is observed, consider validating detection in those systems.
  • For Western blot, a band at approximately 45 kDa is expected; include appropriate molecular weight markers and loading controls.
  • Validate the antibody in physiologically relevant sample types, such as human peripheral blood monocytes or neutrophils.
  • For ELISA applications, optimize antibody dilution and consider appropriate blocking and washing conditions for robust signal detection.
  • When detecting phosphorylated forms, note that activation-dependent phosphorylation may affect band shift or signal intensity.
