Biological Background: p47phox/NCF1 Function and Localization
- Also known as Neutrophil cytosol factor 1, NCF-47K, Nox organizer 2 (NOXO2), and p47-phox. It is a subunit of the phagocyte NADPH oxidase complex.
- As part of the NADPH oxidase complex, it mediates electron transfer from NADPH to O2 to produce superoxide anion (PubMed:2547247, PubMed:2550933, PubMed:38355798). This superoxide generation is critical for the microbicidal activity of phagocytes. Related references: PMID:2547247, PMID:2550933, PMID:38355798
- Activation of the oxidase involves phosphorylation-dependent binding of NCF1 to the C-terminus of CYBA/p22-phox, promoting translocation of cytosolic subunits to the membrane (PubMed:12732142, PubMed:19801500). Related references: PMID:12732142, PMID:19801500
- The protein contains SH3 and PX domains, which are involved in protein-protein and lipid interactions, facilitating complex assembly.
- Subcellular localization includes cytoplasm, cytosol, and membrane; it is primarily cytosolic and translocates to the membrane upon activation.
- Tissue specificity: Detected in peripheral blood monocytes and neutrophils at the protein level.
- Keywords highlight its association with chronic granulomatous disease (CGD), alternative splicing, phosphoprotein, and SH3 domain.
- It plays a role in host defense, with mutations linked to CGD, an immunodeficiency characterized by recurrent infections.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 1-390, covering the entire protein sequence. This may enable detection of multiple isoforms if present.
- For Western blot, the expected molecular weight is approximately 45 kDa; consider verifying in relevant lysates from human, mouse, or rat monocytes/neutrophils.
- For immunofluorescence, permeabilization may be required to detect cytosolic p47phox; validate signal in activated vs. resting phagocytes.
- In immunoprecipitation, the antibody may co-precipitate other NADPH oxidase complex components; confirm with appropriate controls.
- For ELISA, test the antibody as capture or detection reagent in a sandwich assay; optimize concentrations and validate with known standards.
