Biological Background: p38 MAPK Function and Localization

  • Mitogen-activated protein kinase 14 (MAPK14), commonly known as p38 alpha, is a stress-activated serine/threonine kinase belonging to the MAP kinase family. It exists as multiple isoforms including EXIP and MXI2, generated through alternative splicing.

  • Upon activation by extracellular signals such as pro-inflammatory cytokines or physical stress, p38 phosphorylates downstream kinases MSK1/2, which then regulate transcription factors (CREB1, NF-κB, STATs) and histone H3 (PubMed:9687510, PubMed:9792677). It also activates MK2/3 to control mRNA stability via ZFP36 and ELAVL1, and MNK1/2 to phosphorylate EIF4E2 for translation initiation (PubMed:11154262). Experimental Guidance and Technical Tips

  • The immunogen (amino acids 261–360 of human p38) is conserved across human, mouse, and rat, consistent with the antibody’s cross-reactivity. Verify by sequence alignment before extending to other species.
  • For Western blotting, use denaturing reducing conditions and a starting antibody dilution range of 1:500–1:2000; optimize with appropriate positive controls (e.g., HeLa, Jurkat).
  • For IHC-P, antigen retrieval (e.g., citrate buffer, pH 6.0) may be needed; validate staining specificity with blocking peptide or knockout tissue controls.
  • For IF/ICC, fix with 4% PFA and permeabilize with 0.1% Triton X-100; co-stain with nuclear/cytoplasmic markers.
  • For IP, use 1–2 µg antibody per 500 µg protein lysate and capture with Protein A/G beads.
  • For ELISA, the antibody may be used as capture or detection reagent; test pairing with a matched monoclonal antibody.
