Biological Background: p38 MAPK (MAPK14) Function and Localization

  • MAPK14 is a serine/threonine kinase and a member of the p38 MAP kinase family. It is also known as p38 alpha, CSBP, SAPK2A, and has isoforms MXI2 and EXIP. The protein undergoes phosphorylation and acetylation and is involved in stress responses, inflammation, apoptosis, and transcriptional regulation.

  • As a core component of the MAP kinase pathway, p38 MAPK is activated by extracellular stimuli such as pro-inflammatory cytokines and physical stress. It is estimated to phosphorylate 200–300 substrates, orchestrating broad cellular responses.

  • Downstream kinases MSK1 (RPS6KA5) and MSK2 (RPS6KA4) are directly activated by p38 MAPK and phosphorylate transcription factors CREB1, ATF1, NF-kappa-B RELA, STAT1/3, as well as histone H3 and HMGN1, facilitating rapid immediate-early gene induction and chromatin remodeling.
