Biological Background: P2RX7 Function and Localization
- P2RX7 (P2X purinoceptor 7, also known as P2Z receptor or ATP receptor) is a ligand-gated nonselective cation channel belonging to the P2X receptor family.
- Channel activation by high concentrations of extracellular ATP triggers the influx of Na⁺ and Ca²⁺ and efflux of K⁺, and upon prolonged stimulation forms a large pore permeable to molecules up to ~900 Da.
Related references: PMID:17483156 PMID:25281740 PMID:9038151 PMID:20453110 PMID:28235784 - In immune cells, P2RX7 functions as a danger sensor for extracellular ATP, promoting NLRP3 inflammasome assembly and the release of pro-inflammatory cytokines IL-1β and IL-18.
Related references: PMID:26877061 - P2RX7 engagement can trigger programmed cell death pathways, including apoptosis and autophagy.
Related references: PMID:21821797 PMID:23303206 PMID:28326637 - Alternative splicing generates isoforms with distinct functional properties, including an ion channel-only variant that lacks macropore function and non-functional variants.
- The receptor is localized to the plasma membrane and is widely expressed, with the highest levels found in brain and immune tissues.
- Post-translational modifications include N-linked glycosylation, phosphorylation, palmitoylation, and disulfide bond formation, which regulate receptor trafficking and function.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 47–334 of human P2RX7, a region that may include extracellular domains common to multiple isoforms; anticipate detection of several isoform bands in Western blot.
- Due to glycosylation and potential receptor dimerization, the observed molecular weight in WB may exceed the predicted 69 kDa; validate in the appropriate sample system.
- For ELISA, consider optimizing coating buffer pH and blocking conditions to enhance specific signal.
- Given confirmed cross-reactivity with mouse and rat, the antibody can be used for these species, but species-specific validation is recommended.
