Biological Background: Tumor suppressor ARF Function and Localization
- Encoded by the Cdkn2a gene locus, p19ARF (Tumor suppressor ARF) is an alternative reading frame product and a key tumor suppressor, also known as cyclin-dependent kinase inhibitor 2A.
- Induces cell cycle arrest in G1 and G2 phases and acts as a potent tumor suppressor, as demonstrated in mouse models.
Related references: PMID:8521522, PMID:9393858, PMID:15601844, PMID:17936562 - Binds to MDM2 and sequesters it in the nucleolus, blocking MDM2-mediated p53 degradation and thereby enhancing p53-dependent transactivation and apoptosis.
Related references: PMID:10359817, PMID:9529248 - Also triggers G2 arrest and apoptosis independently of p53 by preventing the activation of cyclin B1/CDC2 complexes.
Related references: PMID:15361884 - Localizes mainly to the nucleolus and nucleoplasm, with a mitochondrial isoform that may regulate autophagy and caspase-independent cell death.
- Binds transcription termination factor TTF1 and prevents its nucleolar localization, thereby inhibiting ribosome biogenesis.
Related references: PMID:20513429 - Down-regulates BCL6-induced transcriptional repression and interacts with various proteins including E2F1, MYC, TOP1, and NPM1 to modulate transcription, rRNA processing, and ubiquitination pathways.

Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the N-terminal region (amino acids 1-100) of mouse p19ARF. This region shares homology with human ARF, supporting observed cross-reactivity.
- For Western blot analysis, consider using nuclear or nucleolar-enriched lysates, as p19ARF predominantly localizes to these compartments. A band around 19 kDa is expected.
- ELISA performance may be assessed by peptide competition to confirm antibody specificity.
- When working with p19ARF, note that it can be expressed at low levels under normal conditions; induction of cellular stress or use of tumor cell lines may enhance detection.
