Biological Background: P Glycoprotein Function and Localization

  • P-glycoprotein (ABCB1) is an ATP-dependent translocase that effluxes a broad range of drug substrates and phospholipids across the membrane, playing a key role in multidrug resistance. Related references: PMID:2897240, PMID:35970996, PMID:9038218
  • It catalyzes the outward translocation (flop) of various phospholipids, including phosphatidylcholine and sphingomyelin, from the cytoplasmic to the exoplasmic leaflet of the apical membrane. Related references: PMID:8898203
  • The protein is primarily localized at the apical cell membrane and also detected in the cytoplasm, consistent with a transporter that cycles between compartments.
  • It is highly expressed in tissues with barrier functions such as small intestine, liver, kidney, and brain, influencing drug absorption and distribution.
  • Structurally, it contains two nucleotide-binding domains (NBDs) and twelve transmembrane helices, with functional motifs for ATP-binding and phosphorylation, and is subject to N-linked glycosylation.

  • The immunogen was designed against a region spanning amino acids 393–710 of human P-glycoprotein, which corresponds to an intracellular nucleotide-binding domain (NBD1). This design may facilitate recognition of the native transporter across species.
  • For WB, consider using reducing conditions and adequate protein loading, as P-glycoprotein is a high molecular weight (141 kDa) membrane protein.
  • For ELISA, validate the antibody against recombinant or cell-expressed antigen. Given the reported cross-reactivity with mouse and rat, it may be suitable for preclinical studies.
